Contents Amlodipine besylate
Indications Treatment of HTN & prophylaxis of angina.
Dosage HTN, stable & Prinzmetal's angina Initially, 5 mg once daily. Increase to 10 mg once daily if necessary.
Administration May be taken with or without food
Contraindications Patients w/ cardiogenic shock, recent MI, acute unstable angina & chronic stable angina attack.
Special Precautions Patients w/ hypotension, poor cardiac reserve, heart failure; severe aortic stenosis. Should not be used to treat anginal attack in chronic stable angina. Sudden w/drawal may exacerbate angina. Hepatic impairment. Discontinue if patient experienced ischemic pain.
Adverse Drug Reactions Dizziness, flushing, headache, hypotension, peripheral edema, tachycardia, palpitations, nausea, GI disturbances, increased micturition frequency, lethargy, eye pain, mental depression. Paradoxical increase in ishemic chest pain at the start of treatment. Rashes, fever, abnormalities in liver function (cholestasis), gingival hyperplasia, myalgia, tremor, impotence.
Drug Interactions May enhance antihypertensive effects of other antihypertensives eg ß-blockers, aldesleukin, antipsychotics. Quinidine, carbamazepine, phenytoin, rifampicin, cimetidine & erythromycin.
Pregnancy Category (US FDA) Category C: Either studies in animals have revealed adverse effects on the foetus (teratogenic or embryocidal or other) and there are no controlled studies in women or studies in women and animals are not available. Drugs should be given only if the potential benefit justifies the potential risk to the foetus.
ATC Classification C08CA01 - Amlodipine ; Belongs to the class of selective dihydropyridine derivative calcium-channel blockers with mainly vascular effects. Used in the treatment of cardiovascular diseases.
